The GE AKTA Pilot FPLC System is a robust, automated liquid chromatography platform purpose-built for pilot-scale purification, process development, and small-scale biomanufacturing. At the heart of the system are two high-precision peristaltic pumps: the P-907 system pump and the P-908 sample pump. The P-907 pump features dual modules (A and B) to enable highly accurate binary gradient formation, while the P-908 sample pump is a single-module configuration ideal for sample loading. Both pumps incorporate pressure sensors that continuously monitor pressure levels in the flow path to ensure optimal system performance and prevent overpressure events. The system accommodates flow rates ranging from 4 to 400 ml/min in standard operation, and up to 800 ml/min in dual-pump mode, supporting a wide range of scale-up scenarios. With a maximum operating pressure of 2 MPa (20 bar or 290 psi), the system is compatible with both low- and high-pressure chromatography columns, offering exceptional flexibility for diverse purification strategies.
The AKTA Pilot includes a network of stepper-motor-actuated membrane valves housed within nine individually configured valve blocks, each precisely controlling liquid routing through various stages of the purification process. A sample valve enables flexible inlet selection for loading, while inlet valves guide buffer solutions through a system of pressure sensors and air management components, including an air trap and dual air sensors, to eliminate air bubbles and protect sensitive downstream components such as chromatography columns. These built-in air management features are especially useful for preventing column damage and ensuring reproducibility during scale-up operations.
Downstream of the separation column(s), the system is equipped with an integrated array of detectors that provide continuous in-line monitoring of critical parameters. These include a sanitary pH electrode (range 0 to 14, validated between pH 2 and 12), a conductivity cell (range 1 uS/cm to 999.9 mS/cm), and a UV absorbance detector capable of monitoring up to three wavelengths simultaneously in the 190 to 700 nm range with 1 nm resolution. All detectors are self-calibrating and self-testing to ensure reliable and accurate performance. The pH and conductivity detectors are positioned in close proximity to one another, minimizing dead volume and reducing band broadening, while the conductivity cell includes a temperature sensor to enable real-time temperature compensation for pH readings. The UV cell, placed immediately downstream, enables detection of target molecules with high sensitivity and minimal delay.
System control and data acquisition are managed through UNICORN software, a comprehensive platform that enables method development, gradient control, detector monitoring, and data analysis in a single intuitive interface. The software includes pre-configured templates for common procedures such as media scouting and pH gradient optimization, allowing users to streamline the development of purification protocols. Fraction collection is handled by outlet valves that direct purified product or waste streams based on real-time signal data, enabling highly automated workflows. The system also allows for external device integration and advanced customization for specific bioprocessing needs.
