The Applied Biosystems 7500 Real-Time PCR System is designed for advanced PCR applications, ideal for genetic analysis and molecular diagnostics. This system features a robust 96-well block, supporting reaction volumes from 20 to 100 uL, optimized for precise and efficient thermal cycling. Key characteristics of the system include a temperature range of 4°C to 100°C, ensuring broad flexibility for various assays. The temperature uniformity is maintained at ± 0.5°C, achieved 30 seconds after the cycle starts, with an accuracy of ± 0.25°C, measured three minutes after the cycle begins. This precision is critical for consistent results across all wells.
The system boasts a maximum ramp rate of 2.5°C per second, enabling completion of runs in less than two hours. Its optical setup includes a CCD camera and a halogen lamp for excitation, accompanied by five excitation and emission filters that allow for sensitive multiplex detection of up to five targets per well. With a dynamic range spanning nine logs, the 7500 system can detect a single copy of a template in a 50 uL reaction volume with 99.7% confidence. This sensitivity makes it an indispensable tool for laboratories requiring reliable, high-throughput PCR capabilities. - Thoroughly inspect the display for any dim, flickering, or dead pixels, replacing compromised segments to ensure clear visibility and precise data display
- Verify complete functionality and responsiveness of all buttons and knobs, confirming accurate and reliable input across all controls
- Ensure the machine completes all internal diagnostic checks with zero errors, indicating system readiness and functional integrity
- Confirm the machine accurately identifies attached blocks (96-well, dual 48, etc.), with seamless recognition across different configurations
- Conduct a detailed inspection of internal wiring and all electrical connections, checking for any signs of breaks, disconnections, or tears; replace or repair any underperforming components
- Examine the connector between the block and base for bent or broken pins, replacing as needed to maintain secure, reliable connections
- Calibrate well temperatures with a NIST-traceable thermometer, verifying accuracy and uniformity across all wells to meet specification standards
